After a barren first half at Nsenkyire Sports Arena in Samreboi, the second 45 minutes produced three goals.
Striker Seidu Abubakari broke the deadlock for Samartex 3 minutes after the hour mark.
Francis Gyetuah added the second for the hosts in the 81st minute.
Striker Kofi Kordzi – who returned to the Cities line-up after playing for the Black Galaxies at CHAN 2022 – scored the consolation goal on the 90th-minute mark – his second of the season.
The victory at home sees Samartex leapfrog Legon Cities into 12th position on 23 points at the end of the first round of the league campaign. The Royals have two points less and sit just above the relegation zone – 15th.
